目的探讨早期糖尿病肾病(DN)转化生长因子-β1(TGF-β1)与尿白蛋白的相关性。方法RT-PCR方法测定高糖刺激的系膜细胞内及2型糖尿病小鼠肾小球内TGF-β1mRNA的转录水平;ELISA方法检测DN患者尿TGF-β1和尿白蛋白排泄率(UAER)的变化。结果高糖刺激的系膜细胞内与肥胖db/db小鼠肾小球内TGF-β1mRNA转录水平与对照组比较均明显增高。糖尿病各组患者尿TGF-β1含量均显著高于正常对照组,且UAER增加呈递增趋势,两者呈正相关。结论TGF-β1与早期DN的发生与发展密切相关,是反映早期糖尿病肾损害的重要因子。 相似文献

目的构建真核表达质粒pcDNA3.1(+)-Vasostatin并检测其在真核细胞内的表达水平。方法将带有信号肽的Vasostatin基因片段克隆至pcDNA3.1(+)真核表达载体上,经酶切鉴定及测序分析证明构建成功后,以脂质体介导法转染293T细胞,通过Westernblot法,检测其在293T细胞内的表达水平。结果所构建的真核表达质粒pcDNA3.1(+)-Va-sostatin转染293T细胞后,在其裂解的上清液中,检测到目的基因的表达。结论已成功构建了pcDNA3.1(+)-Vasostatin表达载体,并在真核细胞中表达了目的蛋白。 相似文献

Resin adsorption technique with XAD-8 and XAD-4 was used to characterize the raw water from Erlong reservoir in Jilin province of China. The NOM chemical composition sequences of four organic fractions in the raw water,from high to low,are fulvic acid (FA) fraction,hydrophilic non-acid (HPINA) fraction,hydrophilic acid (HPIA) fraction,and humic acid (HA) fraction. Experimental results show that FA is the main precursor of THMFP among the four organic fractions. However,HA or hydrophobic acid exhibits the highest chlorination activity in forming THMs. It is also found that the value of FI/DOC or SUVA and the specific THMFP have better positive correlation. It is implied that certain source water has unique nature of NOM and DBPs. 相似文献

为了提高钛/瓷结合强度,通过不同方法对钛表面进行处理,研究了钛表面处理后钛/瓷界面组织及结合强度。试验结果表明,烤瓷前进行钛表面喷砂处理,对增强钛/瓷结合强度的影响并不显著,但对抑制钛/瓷界面裂纹的扩展起到积极作用;钛表面预氧化处理能够在钛表面形成一层均匀的氧化膜,使参与界面反应的TiO2增多,反应层增厚,但预氧化温度对钛/瓷结合强度有重要影响;钛表面磁控溅射Zr中间层能够抑制钛表面生成过厚氧化层,而中间层厚度对钛/瓷结合强度也有较大影响。 相似文献

A core-shell structure TiO2/BaFe12O19 composite nanoparticles that can photodegrade organic pollutants in the dispersion system effectively and can be recycled easily by a magnetic field is reported in this paper. The obtained samples were characterized by energy dispersive X-ray spectroscopy (SEM-EDS), X-ray diffraction (XRD) and transmission electron microscopy (TEM). The TiO2/BaFe12O19 magnetic photocatalyst is composed of two parts: (1) TiO2 shell used for photocatalysis and (2) BaFe12O19 core for separation by the magnetic field. The photocatalytic activity of the as-prepared magnetic photocatalyst increased with increasing the thickness of TiO2 coating layer. On the other hand, the saturation magnetizations of titania-coated BaFe12O19 nanoparticles decreased with increasing thickness of the titania coating, while the coercivity does not show any change after coating. 相似文献

The influence of diverse factors on the supercritical fluid extraction (SFE) with supercritical CO2 (scCO2) of galanthamine from bulbs of Narcissus pseudonarcissus cv. Carlton was investigated. The parameters that were studied were CO2 density (temperature and pressure), flow rate and plant material particle size and pre-treatment. The highest yield (303 μg/g) was achieved by extracting 53–1000 μm particle-size powdered dried bulb material moistened with NH4OH (25%, v/v) at 70 °C, 220 bar (690 kg/m3) for 3 h. Other N. pseudonarcissus alkaloids such as O-methyllycorenine and haemanthamine were also obtained. N. pseudonarcissus alkaloids as free bases are highly soluble in CO2 at a high pH as opposed to the slightly soluble salt form in which they are generally found in plants. Therefore, plant material pre-treatment with a base is an essential step for galanthamine extraction. Scanning electron microscope (SEM) results also revealed that the desorption of N. pseudonarcissus alkaloids from the plant material rather than the solubility of the alkaloids in the scCO2 plays a major role in this scCO2 extraction. Protein tyrosine phosphatases(PTPs) are crucial regulators of signal transduction. Among them,PTP-MEG2 is an intracellular enzyme of 593 amino acid residues with a putative lipid-binding domain at the N-terminus. In the present study, we cloned the full-length form of the enzyme and expressed it in E. coli cells as a 6xHis-tagged protein. The majority of the expressed enzyme was found in the inclusion body of E. coli cell extracts.Upon extraction with a buffer containing urea, the recombinant enzyme was purified to near homogeneity on a single Ni-NTA-agarose column. This procedure resulted in the production of over 100 mg of purified recombinant PTP-MEG2 from 1 L E. coli cell culture. The purified protein displayed a single polypeptide band with expected molecular size on SDS-polyacrylamide gel electrophoresis under reducing conditions. Isolated under denatured conditions in urea, the purified enzyme was re-natured by dialyzing against a refolding buffer. The re-natured enzyme effectively dephosphorylated the common PTP substrate para-nitrophenylphosphate with a specific activity of 2000units/mg. Meanwhile, the denatured enzyme was used to immunize a rabbit to produce antibodies. The resulting antiserum had extremely high sensitivity and specificity. When used for Western blot analysis, the anti-serum revealed a wide expression of PTP-MEG2 in many tissues of mice. Together, we developed a highly effective way to purify a large amount of PTP-MEG2 and generated highly sensitive antibodies that can specifically detect endogenous expression of the enzyme in tissues. 相似文献
